Streptolysin O (SLO) is a haemolytic exotoxin that belongs to a large family of cholesterol-dependent cytolysins (CDCs) . CDCs are secreted, monomeric proteins that bind to eukaryotic cells and promote cytolysis through the formation of transmembrane pores . CDC binding requires the presence of high concentrations of soluble cholesterol within the target cell membrane. Following binding the membrane-associated monomers diffuse laterally and polymerize to form a pre-pore oligomer of defined size . Subsequent changes in the tertiary structure of the pre-pore molecule result in the formation of a transmembrane β-barrel which is driven through the cell membrane, facilitating osmotic lysis of the target cell . Species-specific transformation events have resulted in the formation of several different CDC isoforms that mediate adjunct pathogenic interactions in addition to transmembrane pore formation
